Profile - Activity Help Wanted

Multiplexed, trackable CRISPR-based genome engineering for optimization of heterologous protein expression and activity in E. coli

Introduction Heterologous protein expression underpins many advancements in modern molecular biology, including protein engineering, production of bioindustrial enzymes and therapeutics, and structure-function discovery. Heterologous proteins are commonly expressed from plasmids to facilitate cloning and subsequent genetic manipulation. However, plasmid-based expression has many drawbacks, including reduced stability, population drift, and limited control over copy number variations. Combining CRISPR editing tools (1, 2) and advancements in gene sequence engineering (3, 4), heterologous gene integration into chromosomal DNA has become more accessible, offering genomic stability with consistent, high expression (5). These advances facilitate in-depth examination of sequence-function relationships and systematic control of protein activity from the genome. Thorough interrogation of gene sequence variants remains difficult, even across a single locus. To do this efficiently and cost-effectively, libraries of genetic edits must be designed and generated in multiplex, which necessitates trackability of designs to connect phenotype to genotype. An innovative genome engineering technology - the Onyx™ Digital Genome Engineering Platform - performs genome-wide and trackable CRISPR editing at scale in an automated benchtop device. This approach significantly reduces the time and resources spent on constructing variant libraries, as demonstrated in this app note. Here we used Inscripta’s open-sourced MAD7TM CRISPR nuclease to integrate a green fluorescent protein (GFP) gene into the E. coli MG1655 chromosome. We then used the Onyx platform to design and generate GFP site-saturation libraries, with 723 designs targeting functional residues intended to modify fluorescence intensity and spectral characteristics (Figure 1A). Additionally, we generated a library of 8,284 distinct promoter and ribosome binding site (RBS) sequences extracted from the E. coli genome, inserted it 5’ of the GFP gene and screened this library to quantify the strength of native E. coli promoters and RBS sites in a massively parallel experiment.

Pumpkin Pie Foot Scrub

Pumpkins are a nutrient-rich winter squash that offer an array of benefits for the skin. When blended into a smooth pulp, also known as a pure, pumpkins transform into a creamy consistency that can be easily applied to the body. Pumpkin puree, when accompanied with Dead Sea salt, extra virgin olive oil, vegetable glycerin, and cinnamon powder, creates a foot treatment that sloughs away dead skin and nourishes the cracked and hardened skin often found on the bottoms and sides of our feet.

The wonderful thing about this DIY scrub, even if you are a complete stranger to the kitchen, is that you can effortlessly make a beautiful recipe crafted from seasonal ingredients that is just as scrumptious as any pastry dessert.

YOU WILL NEED

  • Bowl
  • 1 cup Dead Sea salt
  • 1/4 cup organic extra virgin olive oil
  • 1 TB. organic pumpkin puree
  • 1 tsp. organic vegetable glycerin
  • 1 tsp. organic cinnamon powder
  • Spoon/Whisk
  • Airtight container
HOW TO MAKE

In a bowl, combine the Dead Sea salt, organic extra virgin olive oil, organic pumpkin puree, organic vegetable glycerin, and organic cinnamon powder. Stir with a spoon or whisk to combine the ingredients. Transfer to a sterilized airtight  container, and label the scrub with the contents and date of creation. you can store the scrub in the refrigerator for up to one week. To use, simply dampen your feet with water and place a large spoonful of the pumpkin pie foot scrub onto each foot. Scrub your feet using gentle circular motions, focusing on areas where your skin may be cracked or dry. Rinse off with warm water.

ABOUT GOVFLEX

It all started in 2014 when a small group of experienced government contractor execs set out to improve the way freelancers and businesses connected and worked together.

At the time we focused on the proposal process to connect proposal managers, pricing experts, graphic artists, and others. So we initially set course as Government Proposal Solutions Inc., GovProp.

We had each operated our own government sector contracting companies or consulting businesses and shared the same observations. Contractors often missed opportunities because they didn’t know how to find the right talent at the right time. Many used antiquated “Rolodex-like” hard copy listings, word of mouth or raw search methods to find help. Setting up consulting agreements and related paperwork was time-consuming and outdated.

Freelance experts and consultants were wrestling with similar problems. Finding opportunities was time-consuming and costly. Paperwork varied from one client to another. Preparing monthly billings was often bureaucratic and worst of all delays in payment often slipped to 60, 90, 120 days, or more. A large amount of freelancer time was filled with uncertainty and non-billable hours.

The GovProp Team researched leading business methods and national trends involving the emergence of freelancers as a growing presence in the U.S. workforce. It became clear that online marketplaces were replacing the old “hunt and peck” methods of finding and working with freelancers. International companies like UpWork and Fiverr were growing exponentially. However, none were experienced in the unique dimensions of the government sector. We knew this would be our niche.

So we pivoted to become GovFlex – The Government Freelance Exchange. As the numbers of freelancers approached the 57 million mark in the U.S. alone, we found a growing trend toward reliance on freelancers as a key element of the Future of Work. About the same time, we found an increasing demand for experienced government sector talent in virtually every technical, administrative and management skill set – not just for pre-award work, but for the entire acquisition cycle including post-contract award delivery of services. Matching freelancers with those who need them remains our primary focus.

As we continue to grow we are responding to the needs of our community of practice. With a focus on quality, we work to enhance our user’s experience, increase reference materials and content in our resource library, expand the GovFlex Academy Pro Shop to provide market research, helpful tools, templates, checklists, and training. Festive Room Spray

Essential oil room sprays are one of the simplest luxuries you can make at home, and they can easily be sprayed in areas prone to odors, such as kitchens, bathrooms, and laundry rooms. Homemade room sprays are an affordable alternative to purchasing a diffuser and also a great way to enjoy the aroma therapeutic benefits of essential oils.

Making such a spray is simple and only requires three ingredients: Vodka, distilled water, and your favorite essential oils. you will also need a dark-colored glass spray bottle for dispensing. The dark color of the glass will prevent light and UV rays from breaking down the essential oils. This recipe is customizable, and we include a variety of lovely scent blends, such as mulled cider, sugar cookie, peppermint hot cocoa, pumpkin spice, and cedar spice. It doesn't matter how clean your house is --- sometimes odors happen. Essential oils are a great ways to lightly fragrance your home without dangerous chemicals.

Mulled Orange Cider

  • 15 drops cinnamon bark essential oil
  • 10 drops orange / sweet orange essential oil
  • 5 drops clove essential oil
Sugar Cookie
  • 10 drops bitter almond essential oil
  • 2-3 drops cinnamon bark essential oil
  • 2-3 drops sweet orange essential oil
Peppermint Hot Cocoa
  • 15 drop cacao absolute
  • 7 drops peppermint essential oil
Pumpkin Spice
  • 15 drops copaiba resin
  • 15 drops vanilla oleoresin
  • 5 drops  cardamom essential oil
  • 2 drops cinnamon bark essential oil
  • 2 drop ginger essential oil
  • 2 drop nutmeg essential oil
  • 2 drops clovebud essential oil
YOU WILL NEED

Mason jar
2 oz. vodka
30-40 drops essential oil blend
1/8 tsp. solubilizer: Mulse D9 (Poly Suga) (optional)
2 oz. distilled water
Glass spray bottle: 4-oz.
Funnel: small

HOW TO MAKE

Start with clean, sterilized supplies. In a mason jar, combine the vodka, essential oil blend, and solubilizer, if desired. Secure the lid on the jar, and shake well to combine. Add the distilled water, and shake again to combine. Transfer the mixture to a glass spray bottle using a small funnel. Shake the room spray again prior to spraying to the essential oils remain distributed in the product.

NOTES
  • Essential oils don't mix into water on their own, but they will combine more easily when diluted with vodka first. If you aren't using a solubilizer, be sure to add the distilled water after shaking the essential oils into the vodka.

  • Vodka helps the holiday scents linger longer and will act as a preservative for the room spray.

  • Don't substitute witch hazel for vodka; it doesn't contain enough alcohol to be effective.

  • Dark-colored essential oils, such as vanilla oleoresin, are more likely to stain fabrics, rugs and carpets. Take care not to spray this recipe on or near wood furniture. 

Arnica Salve

With season and temperature changes approaching, our bodies can become prone to aches and pains during the cooler months. We developed an all-natural arnica healing salve to help prepare you and your body against the stress of headaches and joint and muscle pain. This salve contains the goodness of botanical and oil; it will not only help nourish your body but also your mind. We chose peppermint essential oil due to its refreshing and cooling sensation; however, you can choose lavender for calming effects or sage for its antibacterial properties.

YOU WILL NEED

  • Glass jars
  • 1 tsp. turmeric
  • Dried calendula flowers
  • Dried St. john's wort
  • Dried arnica flowers
  • Carrier oil of choice
  • Cheesecloth / Mesh strainer
  • Double boiler
  • 2 TB. coconut oil
  • 2 TB. Shea / cocoa butter
  • 2 TB. beeswax / vegan candelilla wax
  • 10-15 drops essential oils of choice
  • Glass jars / Metal tins
HOW TO MAKE

In a glass jar, combine the turmeric and your desired amount of dried calendula, St. John's wort, and arnica. Pour over the liquid carrier oil of choice, such as olive oil, and fill to the top. Tightly close the jar and allow to infuse in a sunny Window for four weeks. Strain the liquid into a new jar using cheesecloth or a mesh strainer, and discard the botanicals. In a double boiler, combine 1/2 cup of the infused oil, the coconut oil, shea or cocoa butter, and beeswax or vegan candelilla was on low to medium heat, and stir until completely melted.
